Objective: Investigate the efficacy of Bacillus-based probiotics supplemented at two different levels to modulate the productive performance, egg quality, tibia traits, and specific cecal bacteria counts of Hy-Line Brown layers from 25 to 37 weeks of age. Methods: A total of 216 twenty-five-week-old hens were randomly distributed into 3 experimental diets with 12 replicates of 6 birds per cage. Diets included basal diet supplemented with 0 (CON), 3 × 108 (PRO1), or 3 × 109 (PRO2) CFU of the test probiotic containing Bacillus subtilis PB6, Bacillus subtilis FXA, and Bacillus licheniformis G3 per kilogram of feed. Results: Improved egg weights and mass at 29 weeks; and feed intake at 31 weeks (p < 0.10) were noticed with the probiotic-supplemented PRO1 and PRO2 diets. Considering egg quality, the shell thickness, Haugh units, and yolk color were improved; but yolk cholesterol was lowered (p < 0.05) with PRO1 and PRO2 diets at 29 weeks. At both 33 and 37 weeks, the egg-breaking strength, shell color and thickness, albumen height, Haugh units, and yolk color were improved; but yolk cholesterol was similarly lowered (p < 0.05) with the PRO1 and PRO2 diets. Improved tibia Ca, ash, weights, and density; and raised cecal counts of Bifidobacteria and Lactobacilli (p < 0.05) were noticed with PRO1 and PRO2 diets. Improved tibia P but reduced Clostridia counts (p < 0.10) were also observed with the PRO1 and PRO2 diets. Conclusion: Probiotic supplementation of Bacillus subtilis PB6, Bacillus subtilis FXA, and Bacillus licheniformis G3 at 3 ×108 CFU/kg of feed is adequate to significantly improve egg quality, lower yolk cholesterol, enhance several tibia traits, and raise the populations of beneficial cecal bacteria. Modest improvements in several productive parameters and tibia P but reduced Clostridia were also observed; and could warrant further investigation of probiotic effects beyond the current test period.

BACKGROUND AND AIMS: Stand-replacing crown fires are the most prevalent type of fire regime in boreal forests in North America. However, a substantial proportion of low-severity fires are found within fire perimeters. Here we aimed to investigate the effects of low-severity fires on the reproductive potential and seedling recruitment in boreal forests stands in between stand-replacing fire events. METHODS: We recorded site and tree characteristics from 149 trees within twelve sites dominated by mature black spruce [Picea mariana (Mill.) B.S.P.] trees in the Northwest Territories, Canada. The presence of fire-scarred trees supported classification of sites as unburned or affected by low-severity fires in recent history. We used non-parametric tests to evaluate differences in site conditions between unburned and low-severity sites, and mixed effect models to evaluate differences in tree age, size, and reproductive traits among unburned trees and trees from low-severity sites. KEY RESULTS: Results showed significantly higher density of dead black spruce trees in low-severity sites, and marginally significant higher presence of permafrost. Trees from low-severity fire sites were significantly older, exhibited significantly lower tree growth, and showed a tendency towards a higher probability of cone presence and percentage of open cones compared to trees from unburned sites. Surviving fire-scarred trees affected by more recent low-severity fires showed a tendency towards higher probability of cone presence and cone production. Density of black spruce seedlings significantly increased with recent low-severity fires. CONCLUSIONS: Trees in low-severity sites appeared to have escaped mortality from up to three fires, as indicated by fire scar records and their older ages. Shallow permafrost at low-severity sites may cause lower flammability, allowing areas to act as fire refugia. Low-severity surface fires temporarily enhanced the reproductive capacity of surviving trees and the density of seedlings, likely as a stress response to the fire event.

Introduction: The interpersonal behavior questionnaire (IBQ) is an instrument that measures support and thwarting interpersonal behaviors based on the self-determination theory (SDT). The aim of this work was to adapt the IBQ to the Spanish spoken in Mexico and to examine its psychometric properties (structural validity, discriminant validity, composite reliability, factorial invariance, and nomological validity) in a sample of athletes. Methods: For this purpose, 472 athletes (average age 17.15 years; SD = 1.47) completed a question booklet. Results and discussion: Confirmatory factor analysis supported the structure of six related factors, three factors of behaviors that support autonomy, competence, and relatedness, and three factors of behaviors that thwarting them. The internal consistency of each factor was also supported, as well as the average variance extracted. However, the discriminant validity between the factors of competence and relatedness in their dimensions of support, on the one hand, and thwarting, on the other, is questioned. Factorial invariance was confirmed across gender (men and women) and sport type (individual and team). Nomological validity is in accordance with theory and empirical literature. More studies of the IBQ in sport are necessary to see if these results are a fortuitous product or if they manifest themselves consistently.

The present work delves into the feasibility of employing a novel structured sorbent referred to as GFAD (Guefoam Adsorption Device) for the determination of volatile organic compounds (VOCs) in liquid samples. The chosen method has been static headspace sorptive extraction-thermal desorption gas chromatography mass spectrometry (HSSE-TD-GC-MS). The GFAD comprises an aluminum cellular material with a distinct replication structure and a solid guest phase consisting of activated carbon particles dispersed within the cavities of the cellular aluminum. The extensive specific surface area, robustness, and exceptional thermal conductivity of this pioneering material offer distinct advantages over commercially available polydimethylsiloxane-based Twister® devices. Therefore, the trapping efficiency for volatile organic compounds is enhanced, and it is possible to perform the analysis of concentrated samples. According to computational simulations, it has been demonstrated that GFAD has a high heat conductivity. As a result, the desorption efficiency is improved, and minimal temperature gradients are generated throughout the GFAD during the heating process. Besides, the energy consumption is significantly lowered, thus aligning with environmentally conscientious and sustainable analytical practices.The experimental results give a proof of the suitability of the GFAD for determining gaseous compounds in liquid samples through HSSE-TD-GC-MS. For volatile species, the new material provides higher peak areas and lower limits of detection than a commercially available Twister® device. Furthermore, the GFAD is reusable, its adsorbing properties remaining unchanged during, at least, 100 consecutive analyses. In addition, unlike to the Twister®, no intense siloxane peaks are observed in the chromatograms obtained with the GFAD. The feasibility of qualitative and semi-quantitative analysis with the new accessory has been demonstrated with both standards and a cereal bioethanol real sample.

BACKGROUND: We report the final results of the randomized phase 2 FIGHT trial that evaluated bemarituzumab, a humanized monoclonal antibody selective for fibroblast growth factor receptor 2b (FGFR2b), plus mFOLFOX6 in patients with FGFR2b-positive (2 + /3 + membranous staining by immunohistochemistry), HER-2-negative gastric or gastroesophageal junction cancer (GC). METHODS: Patients received bemarituzumab (15 mg/kg) or placebo once every 2 weeks with an additional bemarituzumab (7.5 mg/kg) or placebo dose on cycle 1 day 8. All patients received mFOLFOX6. The primary endpoint was investigator-assessed progression-free survival (PFS). Secondary endpoints included overall survival (OS), objective response rate, and safety. Efficacy was evaluated after a minimum follow-up of 24 months. RESULTS: In the bemarituzumab-mFOLFOX6 (N = 77) and placebo-mFOLFOX6 (N = 78) arms, respectively, 59.7% and 66.7% of patients were FGFR2b-positive in ≥ 10% of tumor cells. The median PFS (95% confidence interval [CI]) was 9.5 months (7.3-13.7) with bemarituzumab-mFOLFOX6 and 7.4 months (5.7-8.4) with placebo-mFOLFOX6 (hazard ratio [HR], 0.72; 95% CI 0.49-1.08); median OS (95% CI) was 19.2 (13.6-24.2) and 13.5 (9.3-15.9) months, respectively (HR 0.77; 95% CI 0.52-1.14). Observed efficacy in FGFR2b-positive GC in ≥ 10% of tumor cells was: PFS: HR 0.43 (95% CI 0.26-0.73); OS: HR 0.52 (95% CI 0.31-0.85). No new safety findings were reported. CONCLUSIONS: In FGFR2b-positive advanced GC, the combination of bemarituzumab-mFOLFOX6 led to numerically longer median PFS and OS compared with mFOLFOX6 alone. Efficacy was more pronounced with FGFR2b overexpression in ≥ 10% of tumor cells. Confirmatory phase 3 trials are ongoing (NCT05052801, NCT05111626). CLINICAL TRIAL REGISTRATION: NCT03694522.

Background: A heterogeneous geographic distribution of childhood acute lymphoblastic leukemia (ALL) cases has been described, possibly, related to the presence of different environmental factors. The aim of the present study was to explore the geographical distribution of childhood ALL cases in Greater Mexico City (GMC). Methods: A population-based case-control study was conducted. Children <18 years old, newly diagnosed with ALL and residents of GMC were included. Controls were patients without leukemia recruited from second-level public hospitals, frequency-matched by sex, age, and health institution with the cases. The residence address where the patients lived during the last year before diagnosis (cases) or the interview (controls) was used for geolocation. Kulldorff's spatial scan statistic was used to detect spatial clusters (SCs). Relative risks (RR), associated p-value and number of cases included for each cluster were obtained. Results: A total of 1054 cases with ALL were analyzed. Of these, 408 (38.7%) were distributed across eight SCs detected. A relative risk of 1.61 (p<0.0001) was observed for the main cluster. Similar results were noted for the remaining seven ones. Additionally, a proximity between SCs, electrical installations and petrochemical facilities was observed. Conclusions: The identification of SCs in certain regions of GMC suggest the possible role of environmental factors in the etiology of childhood ALL.

OBJECTIVE: This review aims to analyse the effectiveness of body weight-supported gait training for improving gait and balance in patients with motor-incomplete spinal cord injuries. METHOD: Relevant articles were systematically searched in electronic databases to identify randomised controlled trials of body weight-supported gait training (either with methods of robotic, manual, and functional electrical stimulation assistance) versus conventional physical therapy or no intervention. Subjects were >16 years-old with motor-incomplete spinal cord injury (AIS C or D). Primary outcomes were gait-related parameters (functionality, endurance, and speed) and balance. Quality of life was included as a secondary outcome. Articles were selected up to 31 December 2023. RESULTS: Fifteen studies met the inclusion criteria (n = 673). Nine studies used robotic assistance, four trials performed manual assistance, one study functional electrical stimulation assistance, and one trial performed the intervention without guidance. Robot-assisted body weight-supported gait training improved walking functionality (SMD = 1.74, CI 95%: 1.09 to 2.39), walking endurance (MD = 26.59 m, CI 95% = 22.87 to 30.31), and balance (SMD = 0.63, CI 95% = 0.24 to 1.02). CONCLUSIONS: Body weight-supported gait training is not superior to conventional physiotherapy in gait and balance training in patients with motor-incomplete spinal cord injury. However, body weight-supported gait training with robotic assistance does improve walking functionality, walking endurance, and balance, but not walking speed.

(1) Background: Prosocial behavior aligns with the current societal model, where human values hold greater importance considering cultural, social, and personal variables that may influence the opportunity to benefit others. Hence, the objective of this research was established: to understand how diverse factors influence the values of young people, aiming to promote education and enhance prosocial behavior. (2) Methods: This study is quantitative research employing an empirical-analytical, cross-sectional social research method. A validated instrument was used with a sample of 1702 individuals from the city of Melilla, noteworthy for its multicultural context due to its location in North Africa. (3) Results: Inferential analysis was conducted using multiple linear regression to predict future behaviors, focusing on the factors influencing values. Various models were employed, incorporating twelve variables and four scales: sociability, transcendence, culture, and effects. (4) Conclusions: The results and conclusions suggest the need to enhance affect and sociability, primarily among the most prominent factors.

El objetivo del presente trabajo es diseñar y validar la creación del cuestionario de evaluación de la calidad percibida en la gestión deportiva del futbol femenil. El muestreo fue intencional por conveniencia, con un total de 332 jugadoras y un rango de edad de entre los 18 y 51 años (M= 20.92; DE= 3.59). Se llevaron a cabo análisis de validez de constructo por medio de un análisis factorial exploratorio (AFE) así como un análisis factorial confirmatorio (AFC) seguido de análisis de fiabilidad y varianzamedia extractada. El instrumento creado fue el GEDECAFF. Los resultados de los análisis factoriales mostraron 36 ítems divididos en 4 factores: (1) Interacción con el entrenador, (2) Instalaciones, (3) Servicio de personal e (4) Institucional; los indicadores de pertinencia fueron apropiados y explican el 76.25% de la varianza total. Los índices de ajuste, error y chi cuadrado son adecuados CFI = .90, TLI = .89 y RMSEA = .07, χ2/gl = 2.17. Los resultados muestran que Alfa de Cronbach y Omega de McDonald por factores está por encima de .90. En conclusión, el cuestionario GEDECAFF presenta las propiedades psicométricas satisfactorias y atiende las necesidades de los servicios ofrecidos por los distintos clubes, dando oportunidad a la mejora continua de la calidad del servicio en el futbol femenil.(AU)


The purpose of this study is to design and validate the creation of the questionnaire for the evaluation of theperceived quality in the sports management of women's soccer. The sampling was intentional for convenience, with a total of 332 soccer players and an age range between 18 and 51 years (M = 20.92; SD= 3.59). Construct validity analyses were carried out using an exploratory factor analysis (EFA) as well as a confirmatory factor analysis (CFA) followed by reliability analysis and extracted mean variance. The instrument created was the GEDECAFF. The result of the factor analysis showed 36 items divided into 4factors: (1) Interaction with a coach, (2) Facilities, (3) Personal service, and (4) Institutional; the relevance indicators were adequate, explaining 76.25% of the total variance. The fit and error indices are adequate CFI = .90, TLI = .89, and RMSEA = .07, regarding the χ2/df = 2.17. The results show that Cronbach's Alpha and McDonald's Omega by factors is above .90. In conclusion, the GEDECAFF questionnaire presents the appropriate psychometric properties and meets the needs of the services offered by the different women's soccer clubs, opening an opportunity for continuous improvement in the quality of service in women's soccer.(AU)

As análises de validade de construto foram realizadas por meio de uma análise fatorial exploratória (EFA), bem como uma análise fatorial confirmatória (CFA), seguida de análise de confiabilidade e variância média extraída. Actualmente las instituciones deportivas se enfrentan a nuevos retos, que los impulsan a realizar cambios constantes con la finalidad de introducirse y posicionarse como instituciones competitivas del sector deportivo privado, al mismo tiempo que buscan nuevos enfoques para atraer la atención de antiguos y nuevos consumidores orientados en perspectivas como; la calidad y la fidelidad, por lo cual, el objetivo de este estudio es analizar la percepción de la calidad y fidelidad en instituciones deportivas privadas respecto a la edad, género y tiempo de permanencia de los usuarios. Para dicho análisis se recabó una muestra de 347 voluntarios, pertenecientes a tres clubes deportivos privados (108 mujeres y 239 hombres), a la cual se le realizaron análisis de fiabilidad descriptivos y comparativos. Los resultados en las variables de calidad y fidelidad muestran diferencias significativas con respecto a la edad y género de los consumidores, contrario a lo obtenido en el tiempo de permanencia, donde los resultados no muestran una diferencia importante. Currently, sports institutions are facing new challenges, which drive them to make constant changes in order to introduce and position themselves as competitive institutions in the private sports sector, while seeking new approaches to attract the attention of old and new consumers oriented inperspectives such as; quality and loyalty, therefore, the objective of this study is to analyze the perception of quality and loyalty in private sports institutions with respect to age, gender and time of permanence of the users. For this analysis, a sample of 347 volunteers belonging to three private sports clubs (108 women and 239 men) was collected and subjected to descriptive and comparative reliability analyses. The results for the quality and loyalty variables show significant differences with respecttothe age and gender of the consumers, in contrast to the results obtained for the time spent, where the results do not show a significant difference. Atualmente, as instituições desportivas enfrentam novos desafios, que as impulsionam a realizar constantes mudanças para se apresentarem e se posicionarem como instituições competitivas no setor desportivo privado, ao mesmo tempo em que buscam novas abordagens para atrair a atençãode antigos e novos consumidores orientados em perspectivas como qualidade e fidelização, portanto, o objetivo deste estudo é analisar a perceção de qualidade e fidelização em instituições desportivas privadas em relação à idade, sexo e tempo de permanência dos usuários. Para esta análise, foi recolhida uma amostra de 347 voluntários de três clubes desportivos privados (108 mulheres e 239 homens), que foi submetida a análises descritivas e de fiabilidade comparativa. Os resultados para as variáveis qualidade e lealdade mostram diferenças significativas em relação à idade e ao género dos consumidores, ao contrário dos resultados obtidos para o tempo de permanência, onde os resultados não mostram uma diferença significativa. Potentially toxic elements (PTEs), found as environmental contaminants, have been related to endometriosis disease. In this context, the peritoneal fluid (PF) matrix has been poorly studied despite its importance. PF is the environment in which endometriotic lesions reside and communicate with surrounding tissues including tissues and nerve cells. In this work, our investigation group reports the special case of a peritoneal endometriosis patient presenting elevated lead, nickel, and bismuth levels in PF. This patient reported following a vegetarian diet and no toxic habits or occupational exposure. In conclusion, the elevated levels of PTEs found may result from a vegetarian diet or an unidentified environmental exposure source. This report provides new insights regarding the possible etiology of endometriosis disease and potential biomarkers for its diagnosis in early stages, although additional research is needed.

Reactive oxygen species are frequently associated with various cancers including pancreatic ductal adenocarcinomas (PDACs). Superoxide dismutase 2 (SOD2) is an enzyme that plays an important role in reactive oxygen species (ROS) signaling. Investigating the molecular function and biological functions of SOD2 can help us develop new therapeutic options and uncover new biomarkers for PDAC diagnosis and prognosis. Here, we show that nimbolide (NB), a triterpene limonoid, effectively blocks the growth and metastasis of PDACs by suppressing the expression and activity of SOD2. To identify the role of SOD2 in NB-induced anticancer activity, we used RNA interference to silence and plasmid transfection to overexpress it. Silencing SOD2 significantly reduced the growth and metastatic characteristics like epithelial-to-mesenchymal transition, invasion, migration, and colony-forming capabilities of PDACs, and NB treatment further reduced these characteristics. Conversely, the overexpression of SOD2 enhanced these metastatic characteristics. ROS signaling has a strong feedback mechanism with the PI3K/Akt signaling pathway, which could be mediated through SOD2. Finally, NB treatment to SOD2-overexpressing PDAC xenografts resulted in significant inhibition of tumor growth and metastasis. Overall, this work suggests that NB, a natural and safe phytochemical that silences SOD2 to induce high levels of ROS generation, results in increased apoptosis and reduced growth and progression of PDACs. The role of SOD2 in regulating NB-induced ROS generation presents itself as a therapeutic option for PDACs.

Purpose: Administration of exogenous alpha-1 antitrypsin (AAT) is the only specific therapy for the management of pulmonary morbidity in patients with AAT deficiency. It requires weekly or biweekly intravenous infusions, which may impact patient independence and quality of life. Self-administration of AAT therapy is an alternative to reduce the burden for patients who require AAT therapy. We presented herein experts' recommendations for the implementation of a program for the self-administration of AAT. Methods: This project was conducted using a modified nominal group technique and was undertaken in two online meetings involving the participation of 25 experts: specialists in pulmonology (n=17), nurses (n=5) and hospital pharmacists (n=3). Results: The following issues were discussed, and several recommendations were agreed upon on the following topics: a) patient profile and clinical evaluation, establishing selection criteria that should include clinical as well as social criteria; b) role of health care professionals, suggested roles for specialists in pulmonology, nurses, and hospital pharmacists; c) training by the nurse, including recommendations before initiating the training and the content of the training sessions; and d) logistic issues and follow-up, adherence, and patient support. Conclusion: We expect this proposal to increase awareness of this therapeutic alternative and facilitate the implementation of self-administration programs, thus contributing to optimizing the patient experience with AAT therapy. Further research on the outcomes of these programs, especially from the patient perspective, will also help to improve their design and implementation.

Seventeen extra virgin olive oil samples from Valencian Community (Spain) were submitted to a domestic-frying process (180 °C) during different degradation times (5, 10, 30, 60, 120 min). A dispersive liquid-liquid aerosol phase extraction by using a methanol/water (50:50) extracting solution was used to isolate the polyphenol fraction. Total phenolic content (TPC) was determined, whereas the determination of seven individual target polyphenolic compounds (hydroxytyrosol, tyrosol, oleuropein, vanillic acid, p-coumaric acid, ferulic acid, and vanillin) was carried out by using ultrahigh-performance liquid chromatography coupled to a tandem mass spectrometer. Statistically significant differences in the TPC values were found for Blanqueta and Manzanilla samples from different harvesting years. The domestic-frying process impacted the TPC and the individual phenolic compounds content. Thermal treatment for 2 h gave rise to a 94% decrease in the TPC. A first-order kinetic model was suitable to accurately describe the degradation of the individual phenolic compounds.

Brain signalling pathways involved in subclinical anxiety and depressed mood can be modulated via the gut brain axis (GBA), providing the potential for diet and dietary components to affect mood. We investigated behavioural, physiological and gut microbiome responses to the Lacticaseibacillus rhamnosus strain HN001 (LactoB HN001™), which has been shown to reduce postpartum anxiety and depression, and a milk fat globule membrane-enriched product, Lipid 70 (SurestartTM MFGM Lipid 70), which has been implicated in memory in stress-susceptible Wistar Kyoto rats. We examined behaviour in the open field, elevated plus maze and novel object recognition tests in conjunction with the expression of host genes in neuro-signalling pathways, and we also assessed brain lipidomics. Treatment-induced alterations in the caecal microbiome and short-chain fatty acid (SCFA) profiles were also assessed. Neither ingredient induced behavioural changes or altered the brain lipidome (separately or when combined). However, with regard to brain gene expression, the L. rhamnosus HN001 + Lipid 70 combination produced a synergistic effect, reducing GABAA subunit expression in the amygdala (Gabre, Gat3, Gabrg1) and hippocampus (Gabrd). Treatment with L. rhamnosus HN001 alone altered expression of the metabotropic glutamate receptor (Grm4) in the amygdala but produced only minor changes in gut microbiota composition. In contrast, Lipid 70 alone did not alter brain gene expression but produced a significant shift in the gut microbiota profile. Under the conditions used, there was no observed effect on rat behaviour for the ingredient combination. However, the enhancement of brain gene expression by L. rhamnosus HN001 + Lipid 70 implicates synergistic actions on region-specific neural pathways associated with fear, anxiety, depression and memory. A significant shift in the gut microbiota profile also occurred that was mainly attributable to Lipid 70.

INTRODUCTION: Ixekizumab (IXE) is an IgG4-type monoclonal antibody targeting IL-17A indicated alone or in combination with methotrexate, for the treatment of active psoriatic arthritis (PsA) in adult patients with insufficient response or with intolerance to one or more disease-modifying anti-rheumatic drug (DMARD) therapy. The PRO-STIP study aimed to describe persistence, patient characteristics, treatment patterns, and effectiveness in patients with PsA receiving IXE in a real-world clinical setting in Spain. METHODS: This was an observational, multicentric, retrospective, longitudinal study in adult PsA patients who started IXE between January 2019 and December 2020, with at least 24 weeks of follow-up. A descriptive analysis of patient characteristics and treatment patterns was performed. The primary objective, treatment persistence, was estimated by Kaplan-Meier survival curve. Effectiveness was evaluated by Disease Activity in Psoriatic Arthritis (DAPSA) scores at baseline and at 12 and 24 weeks. RESULTS: Eighty-nine patients met the selection criteria (55.1% women and mean age 51.5 years). The median time from PsA diagnosis to starting IXE was 7.7 years (IQR 3.4-14.6). Prior to IXE, 95.5% patients had been treated with at least one biologic or targeted synthetic DMARD (b/tsDMARD). The observed persistence rates were 95.5%, 84.3% and 68.5% at 24, 48, and 104 weeks, respectively. The median persistence was not reached in the study period (mean persistence, 86.9 [95% CI 80.6-93.2] weeks). Twenty-eight (31.5%) patients discontinued IXE, 19 patients (21.3%) due to loss of effectiveness and two patients (2.2%) due to adverse events. In patients receiving treatment and with available effectiveness assessment (n = 24), DAPSA decreased significantly from baseline 23.7 (95% CI 19.5-27.9) to 14.8 (95% CI 10.5-19.2) at 12 weeks (p = 0.005) and 14.3 (95% CI 11.1-17.4) at 24 weeks (p = 0.004). CONCLUSIONS: PsA patients treated with IXE in a real-world setting show high treatment persistence through 104 weeks and improvements in disease activity after treatment initiation. This suggests that IXE could be an effective treatment for patients with PsA. RETROSPECTIVELY REGISTERED: Date of registration: 25th May 2021.

INTRODUCTION: Acute pancreatitis (AP) is the third most common gastrointestinal disease resulting in hospital admission, with over 70% of AP admissions being mild cases. In the USA, it costs 2.5 billion dollars annually. The most common standard management of mild AP (MAP) still is hospital admission. Patients with MAP usually achieve complete recovery in less than a week and the severity predictor scales are reliable. The aim of this study will be to compare three different strategies for the management of MAP. METHODS/DESIGN: This is a randomised, controlled, three-arm multicentre trial. Patients with MAP will be randomly assigned to group A (outpatient), B (home care) or C (hospital admission). The primary endpoint of the trial will be the treatment failure rate of the outpatient/home care management for patients with MAP compared with that of hospitalised patients. The secondary endpoints will be pain relapse, diet intolerance, hospital readmission, hospital length of stay, need for intensive care unit admission, organ failure, complications, costs and patient satisfaction. The general feasibility, safety and quality checks required for high-quality evidence will be adhered to. ETHICS AND DISSEMINATION: The study (version 3.0, 10/2022) has been approved by the Scientific and Research Ethics Committee of the 'Institut d'Investigació Sanitaria Pere Virgili-IISPV' (093/2022). This study will provide evidence as to whether outpatient/home care is similar to usual management of AP. The conclusions of this study will be published in an open-access journal. Toxic metals found in the environment have been linked to female infertility and gynecological illnesses. Reliable analytical methods, such as inductively coupled plasma tandem mass spectrometry (ICP-MS/MS), are necessary to determine the elemental composition of biological samples. Currently, the multielemental profile of peritoneal fluid (PF) samples has not yet been established. Due to the complexity of the PF matrix, an ICP-MS/MS-based method has been optimized to mitigate matrix effects and spectral interferences. A dilution factor of 1:4 was the best option to mitigate matrix effects while keeping sensitivity at an appropriate level. A collision gas (He) was useful to lower the extent of spectral interferences for 56Fe, 52Cr, 63Cu, and 68Zn. An intermediate validation test was performed to evaluate accuracy, achieving recoveries ranging from 90 to 110%. The method was validated in terms of intermediate precision, reproducibility, and trueness, with an expanded uncertainty lower than 15%. Afterward, it was applied to perform the multielemental analysis of 20 PF samples. The concentrations for major analytes were up to 151 µg L-1. Meanwhile,209Bi, 111Cd, 52Cr, 55Mn, 95Mo, 60Ni, 208Pb, 118Sn, and 51V were present at concentrations included within the 1-10 µg L-1 range, while 59Co and 139La were found at concentrations below 1 µg L-1.
